Abstract Macrophages respond to environmental cues and play a crucial role in host defense, inflammation, tissue homeostasis. Macrophage activation undergoes metabolic transcriptomic reprogramming adapt tailor an appropriate response, regulating innate immunity. Reactive oxygen species (ROS) have been shown pivotal macrophage activation. Nevertheless, the detailed molecular mechanism of how redox signals direct is not fully elucidated. Here, we uncover for lysine acetyltransferase MOF (also known as KAT8) inflammatory via modulating acetylation peroxiredoxin 1 (PRDX1), H 2O 2scavenger. We demonstrate that PRDX1 novel substrate MOF. dynamic reversely regulated by HDAC6 SIRT2. fine-tunes its peroxidase activity, leading modulation intracellular ROS level upon LPS stimulation. Moreover, appears participate LPS-induced regulation 2-mediated signal transduction.
